* Mechanism of action:
& & Isotretinoin is a retinoid, which when administered in pharmacological dosage inhibits
& & sebaceous gland function and keratinization. The exact mechanism of action of
& & isotretinoin is unknown, however, clinical improvement in nodular acne in association
& & with a reduction in sebum secretion. The decrease in sebum secretion is temporary and is
& & related to the dose and duration of treatment with Net Look, and reflects a reduction in
& & sebaceous gland size and an inhibition of sebaceous gland differentiation.
* Indications:
& & Severe forms of acne, especially nodulocystic acne and forms of acne with tendency to scarring.
* Dosage and administration:
& & The recommended dosage range for Net Look is 0.5 to 1.0 mg/kg/day given with food for
& & 15 to 20 weeks. Adult patients whose disease is very severe with scarring or is primary
& & manifested on the trunk may require dose adjustments up to 2.0 mg/kg/day.
& & Concurrent topical treatment: concurrent user of other keratolytic or exfoliative anti-acne
& & drugs or UV phototherapy is not indicated. Sun exposure should be avoided.
* Contraindications:
& & Net Look is contraindicated in pregnancy and lactation.
& & Net Look is teratogenic. Therefore it is contraindicated not only during pregnancy but
& & also in women who could become pregnant during treatment. Also it is contraindicated in
& & renal or hepatic impairment, hypervitaminosis A, severe hyperlipidemia, hypersensitivity
& & to the product and in combination with Tetracycline.
* Precautions:
& & Pretreatment and follow-up liver function tests should be performed until the response to
& & Net Look has been established. Also, pretreatment and follow-up blood lipids should be
& & obtained under fasting conditions.
* Pregnancy and lactation:
& & Net Look is contraindicated in pregnancy and lactation. Attention: pregnancy forbidden! Risk of Malformation!
* To be stored at room temperature (not exceeding 30 degrees C), in dry place.
